E.coli inorganic pyrophosphatase.
Description
E.coli inorganic pyrophosphatase (PPase, EC 3.6.1.1) catalyses the hydrolysis of inorganic pyrophosphate (which inhibits amplification reactions) to form orthophosphate: P2O7-4+H2O-> 2HPO4-2.
Unit definition

1 unit will generate 40 nmol of phosphate per min from pyrophosphate under standard reaction conditions.

Reaction conditions
Reaction buffer is compatible with all known polymerase buffers. Titrating the amount of the enzyme used in standard amplification reaction can result in significant improvements in DNA amplification. use 100-200 mU per reaction.
Storage
Store at -20°C.
Applications
in reactions, where pyrophosphate is accumulated (e.g., RNA synthesis with T7 RNA polymerase and reverse transcriptase, sequencing).

Quality control

assayed for the absence of ATPase activity, specific performance tests, SDS-PAGE purity.
